View SamplesIntercropping is a vital technology in resource-limited agricultural systems with low inputs. Peanut/maize intercropping enhances iron (Fe) nutrition in calcareous soil. Proteomic studies of the differences in peanut leaves, maize leaves and maize roots between intercropping and monocropping systems indicated that peanut/maize intercropping not only improves Fe availability in the rhizosphere but also influences the levels of proteins related to carbon and nitrogen metabolism. Moreover, intercropping may enhance stress resistance in the peanut plant (Xiong et al. 2013b). Although the mechanism and molecular ecological significance of peanut/maize intercropping have been investigated, little is known about the genes and/or gene products in peanut and maize roots that mediate the benefits of intercropping. In the present study, we investigated the transcriptomes of maize roots grown in intercropping and monocropping systems by microarray analysis. The results enabled exploration differentially expressed genes in intercropped maize.

View SamplesTo investigate the function of poplar WRKY23, we generated PtWRKY23-overexpressing and -underexpressing (RNAi) plants. Transgenic plants were inoculated with Melampsora rust or mock-inoculated for assessment of rust-resistance and for gene expression profiling using the poplar Affymetrix GeneChip to study the consequences of PtWRKY23 overexpression and underexpression. Transcriptome analysis of PtWRKY23 overexpressors revealed a significant overlap with the Melampsora-infection response. Transcriptome analysis also indicated that PtWRKY23 affects redox homeostasis and cell wall-related metabolism.

View SamplesThe placenta has shown morphological and functional adaption in Meishan and Yorkshire pig breeds during late gestation. While the vast difference of uterine capacity and conceptus genotype between Western and Chinese pig breeds affects the pattern of placental development. Whether the placenta within Chinese pig breed also has similar adaptive changes is still unknown. So, we measured the weight and area of 80 Chinese indigenous Diannan small-ear pig placentas and further selected 6 placentas from extremely high and low litter size groups (HL and LL) for deep RNA-sequencing to detect the molecular basis relate to the observed placental phenotype difference.

View SamplesArabidopsis ATH1 Genome Array was used to study the expression levels of 22,500 probe-sets representing 24,000 genes. Both wild type and different transgenic lines were allowed to grow for one month under normal condition was taken for expression analysis. These plants were treated with half strength of Hoagland solution supplemented with 200mM NaCl in pots for seven days for salinity treatment. High quality RNA was extracted from the healthy leaf samples using TRI Reagent (Ambion, INC. USA) and pooled from at least four independent stressed and non-stressed samples separately, and treated with DNase-I (QIAGEN GmbH, Germany). RNA cleanup was carried out using RNeasy Plant Mini Kit (QIAGEN GmbH, Germany) and 5µg of total RNA from each sample with three biological replications were reverse-transcribed to double stranded cDNA using the GeneChipR One-Cycle cDNA Synthesis Kit. The biotin-labelled cRNA was made using the GeneChipR IVT Labelling Kit (Affymetrix, CA, USA). Twenty microgram of cRNA samples was fragmented and was hybridized for 16 hours at 45°C to the Affymetrix Arabidopsis ATH1 Genome Array (Santa Clara, CA, USA). After washing and staining with R-phycoerythrin streptavidin in a Fluidics Station, using the GenechipR Fluidics Station 450, the arrays were scanned by the GenechipR 3000 Scanner. The chip images were scanned and extracted using default settings and the .CEL files were generated using Affymetrix GeneChip Command Console (AGCC) Software.<br></br>Atleast two ATH1 Genome Array was used for hybridisation for each group of samples.
