To investigate the consequences of reduced expression of core spliceosome factors (Dib1 and Snu114) Overall design: Yeast strains were produced whereby expression of the genes Dib1 or Snu114 was subject to a Galactose inducable promoter. Expression of Dib1 and Snu114 was then temporaly reduced by culturing strains in the absence of galactose for up to 8h. RNA was extracted at 4h and 8h time points and the expression of both gene regions and intron regions of all genes was quantified by RNA-Seq. Control samples were cultured in Galactose-containing media for 6h and processed identically.
