WT and mutant strains (fob1delta,sir2delta,ubr2delta) of Saccharomyces cerevisiae were grown and replicatively aged in mini-stat aging devices for transcriptional (RNA-seq) and chromatin accessibility (ATAC-seq) profiling. Overall design: Yeast cells were collected during exponential growth and labeled with biotin for retention of mother cells using magnetic streptavidin beads in specialized growth vessels. Aging mothers were sampled over time for collection of RNA and DNA for RNA-seq and ATAC-seq respectively.
