Human glioblastoma stem cells are thought to be extremely important in causing recurrence of this aggressive brain tumour and resistance to therapy. However, little is known of the genes needed for these cells to survive. We are performing a genome-wide CRISPR/cas9 screen for the genes essential for proliferation of these cells in vitro. Such genes will have depletion of their guide RNAs, and these candidates will be validated in downstream in vitro and in vivo experimental work, representing potential novel therapeutic targets.
