The oil-in-water emulsion Adjuvant System 03 (AS03) is one of the few adjuvants used in licensed vaccines. AS03 induces a local and transient inflammatory response that contribute to its adjuvant effect.  This data provides a more comprehensive analysis of early gene expression both at the injection site and in the draining lymph node 2, 4 or 6 hours after immunisation with AS03.

This data provides a more comprehensive analysis of early gene expression both at the injection site and in the draining lymph node 2, 4 or 6 hours after immunisation with AS03.


C57BL/6 mice (6-8 weeks old) were immunised in both hind limbs, in the gastrocnemius muscles and in a volume of 50 µl/muscle with AS03 or PBS. Iliac lymph nodes were identified as draining in setup experiments. Each tissue sample consisted of a pool of tissues of 2 mice (either dLN or muscle).

Total RNA was isolated by homogenizing pooled dLN in Tripure reagent (1 ml/100 mg tissue; Roche Applied Science), extracted with chloroform and purified with the RNeasy Minikit (Qiagen) following manufacturer’s instructions, with DNase treatment of the column to remove possible genomic DNA contamination. RNA was concentrated by ethanol precipitation and quantified by RiboGreen™ (Life Technologies)

1 μg of each RNA sample was used for target preparation using a one-cycle cDNA synthesis kit.

RNA was hybridised to GeneChip Whole Mouse Genome 430 2.0 arrays (Affymetrix) following manufacturers instructions

Data acquisition was performed using GeneChip Operating Software (Affymetrix) and data quality control and normalisation was performed with the R and Bioconductor stats packages.  The "Musc PBS 2H_3" sample was excluded because of QC failure
