Splenocytes from adult (6-10 week old) or neonatal (3 day old) C57BL/6 mice were passed through a 70um filter to generate a single cell suspension and red blood cells were lysed with ammonium chloride solution.  Cells were washed with 2%PBS and then incubated in RPMI1640 media (StemCell Technologies, Vancouver, Canada) containing 10% FCS, L-glutamine, penicillin, streptomycin, and 5x10-5 M 2-mercaptoethanol.  After three hours at 37C to remove the adherent splenocytes, the non-adherent cells were cultured in the presence of 1000 U/ml human recombination IL-2 (PeproTech, Rocky Hill, NJ) for 7 days.  After the cultures the cells were blocked with 2.4G2 hybridoma supernatant and stained with anti-NK1.1-PE and anti-CD3epsilon-FITC (BD-Biosciences, Mississauga, ON).  NK cells were purified by two rounds of cell sorting by a FACs caliber (BD, Mountain View, CA).

Neonatal NK cells (all 3 samples) and adult NK cells (all 3 samples) were grown from bulk splenocytes for 7 days in the presence of 1000 U/ml human recombination IL-2 (PeproTech, Rocky Hill, NJ).
