Human atherosclerotic plaques from internal carotid artery were obtained from patients undergoing carotid endarterectomy due to high-grade stenosis. Plaques were removed en bloc in longitudinal endarterectomy, drained with saline and cut into 5 longitudinal slices. One of these was stored in -80°C until total cellular RNA was extracted by using Trizol® reagent (Invitrogen Life Technologies, Inc. Carlsbad, CA) according to manufacturer's recommandations. Shortly, plaques were homogenized in Trizol® using Ultra-Turrax T25 homogenizer (IKA Laboratorioteknik) and subsequent steps were done according to the routine protocol except for an extra centrifugation before addition of chloroform to get rid of unhomogenized material and the majority of DNA. Total RNA was further purified by silica colums (RNeasy; Qiagen, Valencia, CA) according to the manufacturer's instructions. Quality was controlled by measuring A260/280 by spectrophotometry and the 18S/26S ratio by microcapillary electrophoresis (Bioanalyzer 2100; Agilent Technologies, Palo Alto, CA).
