After 5-6 weeks of growth, six or seven pots with similar plant populations were used for the different treatments.  Pseudomonas syringae pv. glycinea strain R4 and P. syringae pv. tomato strain DC3000 carried the plasmid pVSP61 with no insert or with avrRpt2, avrRpm1, avrPphB or avrRps4 under control of their native promoters (Hinsch and Staskawicz, 1996; Innes et al., 1993; Kunkel et al., 1993; Simonich and Innes, 1995).  Strains were grown at 28oC on NYGA plates with 25 &#956;g/ml kanamycin and 50 &#956;g/ml rifampicin for 2 days and then resuspended in 10 mM MgCl2 within 30 minutes before use and diluted to equivalent optical densities corresponding to a final concentration of 1 X 105 cfu/ml.  For treatments, pots of plants were vacuum infiltrated (at 8:30-8:50 AM) with the above bacterial solutions or with 10 mM MgCl2 (mock inoculation), and plants were then returned to their original growth chambers or growth rooms.  All treatments were performed on three pots of plants from three growth chambers or growth rooms.  Fourteen hours after treatment (10:30-10:50 pm), all leaf rosettes in individual pots were collected together by cutting with a razor at the basal stem, and immediately frozen in liquid N2 and stored at –80 oC.
