The aim of the experiment is provide a reference dataset for placing wheat grain transcriptome experiments in a developmental context. RNA was isolated from whole grain tissue of replicate wheat cv. Hereward plants at 6, 8, 10, 12, 14, 17, 21, 28, 35 and 42 days after anthesis (daa). Also supplied are array data for grain sampled at 14, 21 and 28 daa under control, hot, dry and hot&dry conditions to illustrate the importance of developmental context in interpretation.

Wheat seeds cv Hereward germinated and vernalised for 8 weeks at 4o C. Planted 4 seed per 18cm diameter pot in Dec 2005. Rooting medium: 75% L&P Medium grade peat; 12% Screened sterilised loam; 3% Medium grade vermiculite; 10% Grit(5mm screened, lime free); 3.5kg per m3 Osmocote Exact 3-4 month; 0.5kg per m3 PG mix; Lime to pH 5.5-6.0 and wetting agent. Grown in Glasshouse with controlled temperature and supplementary lighting (16 hour photoperiod) giving 200 to 1500 micromoles m-2 s-1 photosynthetically active photon flux.

Rooting medium: 2:1:2:0.5 vermiculite: sand: gravel: compost mixed with 2kg per m3 Osmocote containing ratio of 15:11:13:2 N:P2O5:K2O:MgO.Grown in ambient tunnel until anthesis then transferred to controlled envrionment cabinets. Environment until maturity: light (16 hour photoperiod) of 700 micromoles m-2 s-1 photosynthetically active photon flux; 70% relative humidity; temperature 23/15oC or 28/20oC; water supply to 100% or 44% of soil field capacity.
